Normal human oral keratinocytes are differentiated into tissues with a non-cornified, buccal phenotype, or a cornified, gingival phenotype (as pictured).
The same cells are used to propagate both tissues.
The epithelium is cultured on a microporous membrane (Millipore Millicell™ CM cell culture inserts) on a plastic disk utilizing serum-free media.
This specimen was fixed in 4% paraformaldehyde.
The culture construct was removed from the well with an 8mm biopsy punch, processed between biopsy pads and bisected into two approximately equal halves prior to paraffin embedding.
